Fermentation design process: Introduction to fermentation sterilization principle and fully automatic sterilization technology

In the fermentation industry, wet heat sterilization is widely used for sterilizing culture media and fermentation equipment. In factories, steam is easily available and easy to control operating conditions, making it a simple, inexpensive, and effective disinfection method. When using moist heat sterilization method to treat culture medium, the heating time is related to the degree of sterilization and the degree of nutrient destruction. The reduction of nutrients will affect the cultivation of bacterial strains and the production of products, so the degree of sterilization in fermentation design and the destruction of nutrients become the main contradictions in sterilization. Proper control of heating time is the key to sterilization.

Proper control of heating time is the key to sterilization.

The main factors affecting the sterilization effect of fermentation design culture medium are: 1. The composition of the culture medium. The oils, sugars, and proteins in the culture medium can usually increase the heat resistance of microorganisms and slow down their thermal death rate. This is mainly because organic matter forms a thin film outside the cells of microorganisms, hindering the introduction of heat. Therefore, it is necessary to increase the sterilization temperature or extend the sterilization time. 2. The pH value of the culture medium. Microorganisms are heat-resistant within the pH range of 6.0-8.0. When pH<6.0, hydrogen ions are highly likely to penetrate into cells, altering their physiological responses and promoting cell death. Therefore, the lower the pH, the shorter the sterilization time required. Generally, microbial growth has certain requirements for the pH of the culture medium. If pH adjustment is not allowed, it is necessary to consider extending the sterilization time or increasing the sterilization temperature appropriately. 3. The physical state of the culture medium. The physical state of the culture medium has a significant impact on sterilization. The sterilization time of solid culture medium is longer than that of liquid culture medium. If the sterilization time of liquid culture medium is 1 hour at 100 degrees, then solid culture medium needs 2-3 hours to achieve the same sterilization effect. The reason is that during sterilization of liquid culture media, heat transfer is achieved through conduction and convection, while solid culture media only have conduction without convection. 4. The nature and quantity of microorganisms in the culture medium. The properties and quantities of bacteria contained in different components of the culture medium are different. The more microorganisms in the culture medium, the longer the sterilization time required to achieve sterility requirements. The bacterial content of natural culture media, especially nutrient rich or spoiled raw materials, is much higher than that of chemical materials, and the sterilization time should be appropriately extended. For culture media containing a high amount of Bacillus and mold, it is necessary to increase the sterilization temperature or extend the sterilization time appropriately.

Introduction to automatic sterilization in fermentation design: Sterilization is an essential process in the fermentation process, which determines the success or failure of subsequent bacterial cultivation. Of course, the sterilization process can also cause a certain degree of damage and destruction to the culture medium. The sterilization process can ensure both the sterilization effect and the freshness of the culture medium. At present, traditional fermentation design equipment in China uses manual operation and three-way steam inlet to sterilize fermentation tanks, and there are significant uncertainties caused by human influence. Even with the same medium formula and fermentation design process, the fermentation yield can vary greatly. Currently, most foreign countries adopt intelligent control and automatic sterilization solutions. It can ensure consistent heating and destruction of the culture medium during the sterilization process, and monitoring of multiple cold points on the equipment can ensure that there are no dead corners in the sterilization process. It can also reduce labor and lower the risk of sterilization failure caused by human uncertainty factors.
